The United States EPA published an advance notice of proposed rulemaking (NPRM) in the 12 December 2019 Federal Register seeking to limit EtO emissions. Over the next couple of years, information was collected and a proposed air toxics rule published in the 13 April 2023 Federal Register. Polypeptide C-terminal bis(2-sulfanylethyl)amides (SEA peptide segments) react with Cys peptide to give a native peptide bond as in NCL. This reaction, which is called SEA Native Peptide Ligation, is a useful variant of native chemical ligation. In making peptide segments that contain an N-terminal cysteine residue, exposure to ketones should be avoided since these may cap the N-terminal cysteine. Do not use protecting groups that release aldehydes or ketones. For the same reason, the use of acetone should be avoided, particularly in washing glassware used for lyophilization. A feature of the native chemical ligation technique is that the product polypeptide chain contains cysteine at the site of ligation. The cysteine at the ligation site can be desulfurized to alanine, thus extending the range of possible ligation sites to include alanine residues. This altered remodeling increases the susceptibility of the local endothelium to damage and reduces its repair efficiency. Consequently, this part of the endothelium has an increased risk of being injured and improperly repaired. Thus, the accumulation of misrepairs of the endothelium is focalized and self-accelerating. In this way, the growth of a plaque is also self-accelerating. Within a part of the arterial wall, the oldest plaque is always the biggest and is the most dangerous one to cause blockage of a local artery.
