Thymosin alpha 1 was identified in 1977 as a component of thymosin fraction 5, a heterogeneous preparation used in early studies of thymic function. Investigators purified the active material and determined its amino acid sequence, which enabled chemical synthesis. Work in the following decades concentrated on T-cell maturation and immune reconstitution in animals and small human cohorts. Early preparations varied in composition, so results from that period are difficult to compare with studies using defined synthetic peptide.
Clinical research has examined the peptide in chronic hepatitis B and C, as a vaccine adjuvant, and in sepsis and oncology settings. Findings across trials are mixed; some report changes in selected immune markers, while others find no clear clinical benefit. Many studies are small and define outcomes differently, which limits comparison. Regulatory approval is confined to a few countries, and the compound is not an approved drug in the United States or most of Europe.
Overall evidence quality varies considerably. A large share of published reports come from single centers, rely on surrogate immunological markers, or lack adequate control groups. Systematic reviews have highlighted this heterogeneity as a barrier to pooling results. Open questions include which patients, if any, might benefit, what treatment duration is appropriate, and whether any effect is independent of standard care. The peptide is often described as an immune modulator rather than a therapy for one disease, which complicates confirmatory trial design.
Laboratory work indicates that the peptide acts on cells of both the innate and adaptive immune systems. Reported effects include signalling through Toll-like receptors on dendritic cells, enhanced T-cell maturation, and increased natural killer cell activity. These actions are described largely from cell-culture and animal experiments, and the precise receptor-level events remain incompletely defined. Studies in humans have generally measured immune markers rather than a single defined molecular target. The resulting picture remains partly descriptive.
Clinical research has examined the peptide in chronic hepatitis B and C, as a vaccine adjuvant, and in sepsis and oncology settings. Results across trials have been mixed, and several studies were small or conducted under differing protocols. Regulatory status varies by country, and the compound is not approved in every jurisdiction where it is studied. Evidence for any single indication should be read with attention to sample size and endpoint choice.

Biologically, the peptide is studied mainly in the context of immune cell development and regulation. It is produced in the thymus and in several other tissues, and it appears to influence the maturation and activity of T cells and other immune populations. Laboratory work describes effects on cytokine production, on the balance between T cell subsets, and on the function of dendritic cells. Much of this evidence comes from cell culture and animal models, so the extent to which the same pathways operate in humans remains an open question.
Clinical interest has centered on chronic viral hepatitis, on immune restoration in various conditions, and on use as an adjuvant intended to improve responses to vaccines. Trials have reported mixed results, and regulatory status differs sharply between countries; in some places it is a prescription product, while elsewhere it is sold without an approved therapeutic indication. Because published studies vary widely in design, population, and endpoints, comparisons across them are difficult and no single conclusion covers the whole literature.
Thymosin alpha-1 is a synthetic 28-amino-acid peptide whose sequence was first identified in extracts of bovine thymus tissue during the 1970s. The chain carries an acetyl group on its N-terminal serine. Its acidic residue content is high, which produces strong water solubility and an isoelectric point well below neutrality. Material supplied for laboratory and clinical use is manufactured by solid-phase peptide synthesis rather than purified from animal tissue. Different salt forms, such as the acetate, alter the counter-ion content without changing the peptide backbone.
Whether the free 28-residue peptide circulates in human tissue remains debated. The best-documented human source is prothymosin alpha, a larger acidic protein that carries the sequence at its N-terminus. Reports of measurable peptide levels in serum and lymphoid tissue exist, yet some of that signal may come from cross-reacting fragments or from the parent protein. Most reviews therefore treat prothymosin alpha as the established human molecule and describe independent circulation of the small peptide as an unresolved question.
==== By RuBisCO activase ==== In plants and some algae, another enzyme, RuBisCO activase (Rca, GO:0046863, P10896), is required to allow the rapid formation of the critical carbamate in the active site of RuBisCO. This is required because ribulose 1,5-bisphosphate (RuBP) binds more strongly to the active sites of RuBisCO when excess carbamate is present, preventing processes from moving forward. In the light, RuBisCO activase promotes the release of the inhibitory (or — in some views — storage) RuBP from the catalytic sites of RuBisCO. Activase is also required in some plants (e.g., tobacco and many beans) because, in darkness, RuBisCO is inhibited (or protected from hydrolysis) by a competitive inhibitor synthesized by these plants, a substrate analog 2-carboxy-D-arabitinol 1-phosphate (CA1P). CA1P binds tightly to the active site of carbamylated RuBisCO and inhibits catalytic activity to an even greater extent. CA1P has also been shown to keep RuBisCO in a conformation that is protected from proteolysis. In the light, RuBisCO activase also promotes the release of CA1P from the catalytic sites. After the CA1P is released from RuBisCO, it is rapidly converted to a non-inhibitory form by a light-activated CA1P-phosphatase. Even without these strong inhibitors, once every several hundred reactions, the normal reactions with carbon dioxide or oxygen are not completed; other inhibitory substrate analogs are still formed in the active site.
The Maillard reaction is responsible for many colors and flavors in foods, such as the browning of various meats when seared or grilled, the browning and umami taste in fried onions and coffee roasting. It contributes to the darkened crust of baked goods, the golden-brown color of French fries and other crisps, browning of malted barley as found in malt whiskey and beer, and the color and taste of dried and condensed milk, dulce de leche, black garlic, chocolate, toasted marshmallows, and roasted peanuts. 6-Acetyl-2,3,4,5-tetrahydropyridine is responsible for the biscuit or cracker-like flavor present in baked goods such as bread, popcorn, and tortilla products. The structurally related compound 2-acetyl-1-pyrroline has a similar smell and also occurs naturally without heating. The compound gives varieties of cooked rice and the herb pandan (Pandanus amaryllifolius) their typical smells. Both compounds have odor thresholds below 0.06 nanograms per liter.
British observers believed the war to be all but over after the capture of the two capitals. However, the Boers had earlier met at the temporary new capital of the Orange Free State, Kroonstad, and planned a guerrilla campaign to hit the British supply and communication lines. The first engagement of this new form of warfare was at Sanna's Post on 31 March where 1,500 Boers under the command of Christiaan de Wet attacked Bloemfontein's waterworks about 37 kilometres (23 mi) east of the city, and ambushed a heavily escorted convoy, which caused 155 British casualties and the capture of seven guns, 117 wagons, and 428 British troops. After the fall of Pretoria, one of the last formal battles was at Diamond Hill on 11–12 June, where Roberts attempted to drive the remnants of the Boer field army under Botha beyond striking distance of Pretoria. Although Roberts drove the Boers from the hill, Botha did not regard it as a defeat, for he inflicted 162 casualties on the British while suffering only around 50 casualties.
=== Disorder in the bound state (fuzzy complexes) === Intrinsically disordered proteins can retain their conformational freedom even when they bind specifically to other proteins. The structural disorder in bound state can be static or dynamic. In fuzzy complexes structural multiplicity is required for function and the manipulation of the bound disordered region changes activity. The conformational ensemble of the complex is modulated via post-translational modifications or protein interactions. Specificity of DNA binding proteins often depends on the length of fuzzy regions, which is varied by alternative splicing. Some fuzzy complexes may exhibit high binding affinity, although other studies showed different affinity values for the same system in a different concentration regime.

==== Protease-specific Arrays ==== A protease specific protein array based on immobilized antibodies designed to capture specific proteases from biological samples offers a step up in analysis of protein levels beyond transcript expression. Capture antibodies spotted to nitrocellulose membranes can bind proteases in complex mixtures which have been pre-incubated and bound by detection antibodies allowing for parallel analysis of relative protease levels. These arrays offer parallelization of protein levels over traditional western blot. Unfortunately, these assays fail to provide insight on enzymatic function for proteases and suffer similar drawbacks to western blots regarding reliable quantification.
=== Italy === Since the Fleischmann and Pons announcement, the Italian national agency for new technologies, energy and sustainable economic development (ENEA) has funded Franco Scaramuzzi's research into whether excess heat can be measured from metals loaded with deuterium gas. Such research is distributed across ENEA departments, CNR laboratories, INFN, universities, and industrial laboratories in Italy, where the group continues to try to achieve reliable reproducibility (i.e. getting the phenomenon to happen in every cell, and inside a certain frame of time). In 2006–2007, the ENEA started a research program which claimed to have found excess power of up to 500 percent, and in 2009, ENEA hosted the 15th cold fusion conference.
Exhibitors from outside Tamil Nadu also echoed similar views. Arora Talkies (Mumbai) described Darbar as the most profitable Tamil film screened at their theatre, while exhibitors in Karnataka reported a favourable audience response. Several theatre owners dismissed circulating reports of losses as “rumours” and emphasised that the film delivered satisfactory returns at the exhibition level.
=== Other symptoms === Breathing difficulties can occur, resulting from neuromyotonic activity of the laryngeal muscles. Laryngeal spasm possibly resulting from neuromyotonia has been described previously, and this highlights that, in patients with unexplained laryngospasm, neuromytonia should be added to the list of differential diagnoses. Studies have shown subtly decreased metabolism on positron emission tomography (PET) and single photon emission computed tomography (SPECT) in the left inferior frontal and left temporal lobes. and or basal ganglia hypermetabolism. Ancillary laboratory tests including MRI and brain biopsy have confirmed temporal lobe involvement. Cranial MRI shows increased signal in the hippocampus. Cerebral spinal fluid (CSF) shows normal protein, glucose, white blood cell, and immunoglobulin G (IgG) levels, but there are weak oligoclonal bands, which are absent in the blood serum. Marked changes in circadian serum levels of neurohormones and increased levels of peripheral neurotransmitters were also observed. The absence of morphological alterations of the brain pathology, the suggestion of diffusion of IgG into the thalamus and striatum, more marked than in the cortex (consistent with effects on the thalamolimbic system) the oligoclonal bands in the CSF and the amelioration after PE all strongly support an antibody-mediated basis for the condition. Raised CSF IgG concentrations and oligoclonal bands have been reported in patients with psychosis.

Trials differ in patient population, dose schedule, background treatment, and the endpoints used to judge success. Many are small and single-center, so random variation can dominate the reported effects.
Authorization is limited to a small number of countries and covers specific indications such as chronic hepatitis B and vaccine adjuvant use. Availability and labelling differ by jurisdiction.
Most reviews describe it as an immunomodulatory agent with an uncertain clinical effect. They generally call for larger, better-controlled trials before firm conclusions are drawn.
It corresponds to the first 28 amino acids of thymosin beta-4, a larger protein found in many tissues. The fragment is acetylated at its N-terminus and is produced synthetically for research and pharmaceutical use. Synthetic and natural forms share the same sequence.
