Everything below concerns Thymalfasin. We keep the language plain, cite what the science says, and separate well-supported claims from open questions.

Clinical interest has centered on chronic viral hepatitis, on immune restoration in various conditions, and on use as an adjuvant intended to improve responses to vaccines. Trials have reported mixed results, and regulatory status differs sharply between countries; in some places it is a prescription product, while elsewhere it is sold without an approved therapeutic indication. Because published studies vary widely in design, population, and endpoints, comparisons across them are difficult and no single conclusion covers the whole literature.
Thymosin alpha 1 is a short peptide of 28 amino acid residues that derives from the amino terminal region of a larger precursor protein known as prothymosin alpha. The peptide carries an acetyl group on its first residue and contains no disulfide bonds or carbohydrate chains. Its sequence is highly conserved across mammalian species, which is one reason laboratories treat it as a molecule with a defined and reproducible structure rather than a variable tissue extract. The name follows an early naming convention for thymus-derived fractions and does not imply that the peptide acts as a hormone in the classical endocrine sense.
Overall evidence quality varies considerably. A large share of published reports come from single centers, rely on surrogate immunological markers, or lack adequate control groups. Systematic reviews have highlighted this heterogeneity as a barrier to pooling results. Open questions include which patients, if any, might benefit, what treatment duration is appropriate, and whether any effect is independent of standard care. The peptide is often described as an immune modulator rather than a therapy for one disease, which complicates confirmatory trial design.
Thymosin alpha 1 was identified in 1977 as a component of thymosin fraction 5, a heterogeneous preparation used in early studies of thymic function. Investigators purified the active material and determined its amino acid sequence, which enabled chemical synthesis. Work in the following decades concentrated on T-cell maturation and immune reconstitution in animals and small human cohorts. Early preparations varied in composition, so results from that period are difficult to compare with studies using defined synthetic peptide.

The peptide was identified during work in the 1970s on thymosin fraction 5, a partially purified extract of calf thymus. Investigators separated that mixture and characterized individual components, one of which they named thymosin alpha-1. The same compound later received the international nonproprietary name thymalfasin. Commercial material is produced by solid-phase peptide synthesis rather than by extraction, so synthetic and natural forms share an identical sequence. Naming conventions vary across the literature, and readers should distinguish the alpha-1 peptide from other thymosins that have unrelated sequences and functions.
Thymosin beta-4 is a separate 43-residue peptide that binds actin and participates in cell migration; it shares no sequence similarity with thymosin alpha-1 despite the common family name. Other preparative materials, such as thymosin fraction 5 and thymopoietin, contain distinct mixtures or peptides. The shared thymosin label reflects the tissue of origin used in early purification, not a common structural core. Treating these molecules as interchangeable is a frequent source of confusion in laboratory reports and in popular summaries alike.
Thymosin alpha-1 is a synthetic peptide of 28 amino acid residues that corresponds to a naturally occurring fragment first isolated from thymus tissue. Its chain is acetylated at the amino terminus, a modification that shields the peptide from rapid cleavage by aminopeptidases. The molecule carries a net negative charge at physiological pH and dissolves freely in water. Researchers classify it as an immune-modulating agent rather than a classical hormone, because it acts on several cell types of both the innate and the adaptive immune system.
Thymosin alpha 1 is approved as a medicine in several countries, including Italy and China, for indications such as chronic hepatitis B and as an immune adjuvant. It is not approved by the United States Food and Drug Administration as a therapeutic product. In research settings the peptide appears in studies of sepsis, vaccine response, and oncology support, often with mixed or inconclusive results. The evidence base is uneven, and reviews note that many trials were small. Regulatory status therefore differs widely between jurisdictions.
Thymosin alpha 1 is a synthetic 28-amino-acid peptide first isolated in 1966 from thymosin fraction 5, a bovine thymus extract. Its chain begins with an acetylated serine residue and ends with asparagine. The native peptide carries a molecular mass near 3,108 daltons. Researchers classify it as an immunomodulatory agent rather than a hormone with a single endocrine target. Early work framed it as a thymus-derived factor that supports T-cell maturation. The synthetic form used in research and clinical products matches the natural sequence.
Lyophilized material is typically treated as a hygroscopic solid that should be brought to room temperature before the container is opened, which limits condensation on the powder. Reconstitution is commonly done with sterile water or a buffered diluent, and gentle mixing is preferred over vigorous agitation to reduce foaming and surface adsorption. Because peptides can bind to plastic and glass, diluents containing a small amount of carrier protein are sometimes used in laboratory work. Working solutions are generally aliquoted and prepared fresh rather than subjected to repeated freezing and thawing.
Stability depends on temperature, pH, and the number of freeze-thaw events the sample has experienced. Freeze-dried material is commonly held at -20 °C or colder, while reconstituted liquid is kept cold and used within a short window. Extreme pH and prolonged light exposure can promote deamidation, oxidation, or aggregation, particularly at asparagine and methionine positions. Adsorption to container walls can lower the measured concentration of a dilute solution even when the peptide molecules themselves remain intact.
It was a serious public-relations problem for Abbott, so the company held interviews and press conferences, at which senior Abbott officials answered questions. The transcripts are archived at [1]. A later study found 3 additional morphs: a metastable polymorph, a trihydrate, and a formamide solvate.
=== Preclinical safety data === Conventional long-term toxicity, reproduction, mutagenicity, and carcinogenicity studies have been performed in laboratory animals. Salmon calcitonin is devoid of embryotoxic, teratogenic, and mutagenic potential. An increased incidence of pituitary adenomas has been reported in rats given synthetic salmon calcitonin for 1 year. This is considered a species-specific effect and of no clinical relevance. Salmon calcitonin does not cross the placental barrier. In lactating animals given calcitonin, suppression of milk production has been observed. Calcitonin is secreted into the milk.
=== Fibromyalgia === Studies have also shown paroxetine "appears to be well-tolerated and improve the overall symptomatology in patients with fibromyalgia", but is less robust in helping with the pain involved.
=== Pharmacokinetics === In non-human primates, the plasma half-life of the prodrug is 20 minutes, with the main metabolite being the nucleoside, GS-441524. Two hours post injection, the main metabolite GS-441524 is present at micromolar concentrations, whilst intact Remdesivir is no longer detectable. Because of this rapid extracellular conversion to the nucleoside GS-441524, some researchers have questioned whether the active nucleotide triphosphate is truly derived from remdesivir pro-drug removal or whether it occurs by GS-441524 phosphorylation, and whether direct administration of GS-441524 would constitute a cheaper and easier to administer COVID‑19 drug compared to remdesivir. The activated nucleotide triphosphate form has sustained intracellular levels in PBMC and presumably in other cells as well.
== Adverse effects == Most common side-effects are gastrointestinal, including diarrhea, nausea, abdominal pain and vomiting. Headache and disturbances in taste also occur. Less common side-effects include palpitations, blurred vision, and rashes. Prolonged QTc intervals may also be caused by telithromycin. Rare but severe side-effects were initially reported in March 2006, involving damage to the liver. Three different incidents were reported: one case of temporary drug-induced hepatitis, one ending in a liver transplant, and one ending in death. In the United States, the FDA's Office of Epidemiology and Surveillance identified 12 cases of acute liver failure, resulting in four deaths, and an additional 23 cases of acute, serious liver injury, among 5.2 million patients taking telithromycin through April 2006. In 2010, a published report described the likely mechanism of action underlying not only the cases of liver failure but also cases of visual disturbances and exacerbations of myasthenia gravis. The study showed that a pyridine moiety that is part of the telithromycin molecule acts as an antagonist on cholinergic receptors located in the neuromuscular junction, the ciliary ganglion of the eye and the vagus nerve innervating the liver. Other macrolides, such as azithromycin and clarithromycin and the fluoroketolide, solithromycin, do not contain the pyridine moiety and do not antagonize these cholinergic receptors significantly.

=== Cyst stage === To endure harsh environmental conditions, trophozoites transform into microbial cysts, spherical, single-layered structures about 7–15 μm in diameter, enclosing a single cell nucleus. Acting as a resilient capsule, the cyst enables the amoeba to withstand adverse circumstances. Factors triggering cyst formation include food scarcity, overcrowding, desiccation, waste accumulation, and cold temperatures. When conditions improve, the amoeba can emerge through the pore or ostiole at the center of the cyst. N. fowleri has been observed to encyst at temperatures below 10 °C (50 °F).
=== Neurodegeneration === CRH has also been shown to promote neurodegeneration, suggesting that CRH1 antagonists may have neuroprotective effects. PC12 cells are derived from the rat adrenal medulla and are extensively used to study neural differentiation. PC12 cells treated with CRH (1-10 nM) showed increased numbers of apoptotic cells and upregulation of the Fas ligand via p38 activation, demonstrating the pro-apoptotic effects of CRH. Administration of antalarmin (10 nM) completely blocked the CRH-induced apoptosis response and inhibited Fas ligand expression.
== History == The company was founded in April 2012 by former Morgan Stanley investment banker Adam Forste, who formed Bay Grove Capital, LLC, through the consolidation of acquired warehousing and logistics companies beginning, in December 2008, with the purchase of Seafreeze from Toyo Susian Kaisha. The company then acquired CityIce, in 2009, and Flint River Services, in 2010, followed, in 2011, by Terminal Freezers of Santa Maria, California, and Richmond Cold Storage, purchased from Atlanta Equity. Lineage was formed with the fifth-largest network of temperature-controlled warehouse facilities in North America in 2012, according to International Association of Refrigerated Warehouses (IARW) rankings, with 40 facilities in eight states, including near international ports at Savannah, Georgia, Seattle, Washington and Norfolk, Virginia; as well as providing warehouse management, consulting, and food processing support. In a March 2021 interview with Bloomberg, Lineage CEO Greg Lehmkuhl said the company is actively preparing its initial public offering (IPO). The company completed its IPO in July 2024 with a listing on the Nasdaq, raising around $4.5 billion, valuing the company at over $18 billion. On April 21, 2024, a fire broke out at the Lineage cold storage facility in Benton County, Washington. The fire burned for 60 days. On June 17, 2026 a large fire broke out at the Lineage cold storage facility in Boyle Heights, Los Angeles, accelerated by the large facility's solar panels encompassing its entire roof, refrigerants, and decaying food within the building.
==== Moisture movement directions for diffusion ==== It is reported that the ratio of the longitudinal to the transverse (radial and tangential) diffusion rates for wood ranges from about 100 at a moisture content of 5%, to 2–4 at a moisture content of 25%. Radial diffusion is somewhat faster than tangential diffusion. Although longitudinal diffusion is most rapid, it is of practical importance only when short pieces are dried. Generally the timber boards are much longer than in width or thickness. For example, a typical size of a green board used for this research was 6 m long, 250 mm in width and 43 mm in thickness. If the boards are quartersawn, then the width will be in the radial direction whereas the thickness will be in tangential direction, and vice versa for plain-sawn boards. Most of the moisture is removed from wood by lateral movement during drying.

==== Needle Injector ==== A needle injector is used when setting the facial features to close the mouth. It implants metal pins into the maxilla and mandible. The pins have wires attached that are twisted together to keep the mouth from opening.
Vanilla planifolia is a species of vanilla orchid native to Mexico, Central America, Colombia, and Brazil. It is one of the primary sources for vanilla flavouring, due to its high vanillin content. Common names include flat-leaved vanilla and West Indian vanilla (which is also used for the Pompona vanilla, V. pompona), though it is often simply referred to as vanilla. It was first scientifically named in 1808. Vanilla planifolia played a large role in the early rural economy throughout Mexico and by the 20th century had become a widely cultivated crop across the world. With the species' population in decline and its habitats being converted to other purposes, the IUCN has assessed Vanilla planifolia as Endangered.
=== Therapeutic evaluation === Several investigations correlate the Bristol stool scale in response to medications or therapies, in fact, in one study was also used to titrate the dose more finely than one drug (colestyramine) in subjects with diarrhoea and faecal incontinence. In a randomised controlled study, the scale was used to study the response to two laxatives: Macrogol (polyethylene glycol) and psyllium (Plantago psyllium and other species of the same genus) of 126 male and female patients for a period of 2 weeks of treatment; failing to show the most rapid response and increased efficiency of the former over the latter. The primary outcomes were the number of weekly bowel movements, stool consistency according to the Bristol stool scale, time to defecation, overall effectiveness, difficulty in defecating, and stool consistency. From 2010, several studies have used the scale as a diagnostic tool validated for recognition and evaluation of response to various treatments, such as probiotics, moxicombustion, laxatives in the elderly, preparing Ayurvedic poly-phytotherapy filed TLPL/AY, psyllium, mesalazine, methylnaltrexone, and oxycodone/naloxone, or to assess the response to physical activity in athletes.
=== Terrestrial plants in aquatic environments === Terrestrial plants may undergo physiological changes when submerged due to flooding. When submerged, new leaf growth has been found to have thinner leaves and thinner cell walls than the leaves on the plant that grew while above water, along with oxygen levels being higher in the portion of the plant grown underwater versus the sections that grew in their terrestrial environment. This is considered a form of phenotypic plasticity as the plant, once submerged, experiences changes in morphology better suited to their new aquatic environment. However, while some terrestrial plants may be able to adapt in the short-term to an aquatic habitat, it may not be possible to reproduce underwater, especially if the plant usually relies on terrestrial pollinators.
chloroplast A type of small, lens-shaped plastid organelle found in the cells of green algae and plants which contains light-sensitive photosynthetic pigments and in which the series of biochemical reactions that comprise photosynthesis takes place. Like mitochondria, chloroplasts are bound by a double membrane, contain their own internal circular DNA molecules from which they direct transcription of a unique set of genes, and replicate independently of the nuclear genome.

The name reflects an early convention for naming thymus-derived fractions. The peptide is characterized and measured as a defined molecule, and it does not operate through a single classical endocrine axis.
Trials differ in patient population, dose schedule, background treatment, and the endpoints used to judge success. Many are small and single-center, so random variation can dominate the reported effects.
Authorization is limited to a small number of countries and covers specific indications such as chronic hepatitis B and vaccine adjuvant use. Availability and labelling differ by jurisdiction.
Most reviews describe it as an immunomodulatory agent with an uncertain clinical effect. They generally call for larger, better-controlled trials before firm conclusions are drawn.
